For the purpose of establishment of the IL 2-producing T cell hybridoma, somatic cell hybridization between spleen cells of BALB/c mouse and T lymphoma cell line BW5147. G. 1.4. was performed. And, after expansion and recloning of the original
clone
(TDl), a stable IL 2 producing hybrid cell line (TDl. 4) which possessel the maximal IL 2 producing capacity with Con A and TPA stimulation was established. Now, the TD1.4 hybrid cell line has been producing the mouse IL 2 for 11 months. The
level
of IL
2 produced from TD1. 4 hybrid cell line was at least 20-fold greater than that of Con Astimulated mouse spleen cell. In addition, to establish the best conditions for IL 2 mass production, important factors which affect the IL 2 productivity were
tested. TPA inhibited the proliferation of TD1.4 cells and diminished the Con A stimulated IL 2 production of TD1.4 cells. TD1.4 cell lines produced more IL 2 when cultured in serum-free DMEM with 10sμg/ml Con A an 1.0×10E6 cells/ml, and
3.0×10E5
cells/cm3 for 24-48 hrs.
